TALEs:识别DNA的有力工具

摘要 植物致病菌黄单胞杆菌可以产生一种通过一连串的重复单元特异识别并激活植物基因启动子的蛋白—TALEs。TALEs与DNA识别的关键在于2个连续的多态性氨基酸可以组成1个TALE-DNA结合密码,每个密码特异结合1个核苷酸。整个TALEs蛋白与双链DNA结合,其重复单元的顺序决定了可以结合的核苷酸序列。用人工设计的TALE 核酸酶(TALENs)可以对包括植物、昆虫和哺乳动物在内的广泛的物种进行目的基因的编辑;而且人工设计的TALEs也可以与不同的功能结构融合,起到包括激活和调节基因表达在内的广泛作用,因此为生物技术和基因治疗方面提供了一个良好的工具。
