Abstract:Objective To investigate Borrelia burgdorferii infection in ticks and rodents captured in Qinghai Province and to identify their genotypes.Methods Ticks were collected by dragging a cloth over the vegetation while rodents were collected by snaps. After species identification, DNA was isolated from ticks and detected by nested PCR targeting 5S-23S rRNA intergenic spacer of Borrelia burgdorferii. Positive samples were further analysed by rest riction f ragment length polymorphism (RFLP) .Results A total of 618 adult ticks of 4 species, and 121 rodents of 2 species were tested for evidence of Borrelia burgdorferii by nested PCR.97 tick samples and 12 rodent samples showed positive results. The positive rate was different between different tick and rodent species, with Haemaphysalis qinghaiensis and Rattus tanezumi having the highest positive rates. The positive rate was 21.66% for Haemaphysalis qinghaiensis and 17.39% for Rattus tanezumi. RFLP analysis indicated that Borrelia burgdorferii in samples belonged to either B. garinii or B. afzelii.Conclusion Borrelia burgdorferii exists in Qinghai Province and is of at least two genotypes.
