Objective To investigate the effects of vitamin C (VC) intervention on immune function in rats exposed to high-temperature. Methods Twenty-four SPF 8-week-old Sprague-Dawley (SD) rats were randomly divided into four groups: normal temperature control (NC), high temperature exposure (HE), HE supplemented with 10 mg/kg VC and 14 mg/kg VC. Two intervention groups were administrated intragastrically with 1.0 ml VC solution at the concentration of 1.0 g/L or 1.4 g/L, and the other groups were given the same amount of distilled water every morning. The rats ran on a treadmill in a normal environment (22±1) ℃ or a heat environment (33±1) ℃ for 30 min in the morning and in the afternoon daily, consecutive six days once a week for four weeks. The rats were sacrificed on the 29th day. Flow cytometry was used to analyze the proportions of CD3+, CD4+, CD8+, CD16+ and CD45+ cells in peripheral blood. The levels of IgA,IgG, IgM, C3, and TNF-α, IL-2, IL-6 in the serum were measured by enzyme-linked immuosorbent assay. Results Compared with the NC group, the percentages of T cells, NK cells, WBC, CD4+/CD8+ ratio in the blood,and the contents of IL-6, IgA, IgG and C3 in the serum were significantly decreased. The IL-2 level was significantly increased in the HE group (all P<0.05). Compared with the HE group, the percentages of T cells and NK cells in the 10 mg/kg VC and 14 mg/kg VC groups, as well as CD4+/CD8+ ratio, the percentage of WBC, the contents of IgA, IgG, IgM, C3, TNF-α and IL-6 in the 14 mg/kg VC groupwere significantly increased (P<0.05). There was no significant difference in the content of IL-2 between two VC supplementation groups and the HE group. Conclusion High temperature exposure has profound effect on both cell-mediated immunity and humoral immunity in rats. The intervention by 14 mg/kg VC can alleviate those negative effects caused by high temperature exposure.
