Protective Effect of Branched-Chain Amino Acid on Muscle Atrophy in Weightlessness-Simulated Rats |

Abstract Objective To investigate the protective effect of branched-chain amino acid(BCAA)on muscle atrophy in weightlessness-simulated rats.Methods Weightlessness was simulated by a tail-suspended model in rats. SD rats were randomly divided into 3 groups: ground control group (GC), tail-suspended control group (TC) and tail-suspended with BCAA intervention group (TBI).All the rats were successively fed with normal diets. The rats in GC and TC groups received pure water, and those in TBI group were given pure water and intervened with BCAA solution via intragastric administration respectively. The biochemical indices of serum and soleus (SOL) were detected after 4 weeks.Results Compared with GC group, the muscle wet weight(MWW) of and the ratio of MWW to body weight of rats in the TC group were significantly decreased(P<0.01),the level of serum BUN and Ub was significantly increased(P<0.05),the level of serum IGF-1、TESTO、SOD、 and GSH-Px was significantly decreased(P<0.05), and the expression of atrogin-1 protein in SOL decreased by 32%. Compared with TC group, the muscle wet weight(MWW) and the ratio of MWW to body weight were significantly increased (P<0.01) in TBI group,so was the serum level of IGF-1、TESTO、Ub, SOD and GSH-Px,,but the serum BUN and the expression of atrogin-1 protein in SOL were decreased,while the level of SOD and GSH-Px was increased significantly(P<0.05). (P<0.05).Conclusion BCAA may promote the synthesis of SOL protein, but inhibit its breakdown to some extent,alleviating SOL atrophy in tail-suspended rats. The mechanism potentially lies in facilitating the secretion of anabolic hormone and antioxidation ability,and inhibiting the expression of atrogin-1 protein.
