ZHENG Fengjiao, FU Yonghang, CHEN Wensi, LIU Nan
Journal of Preventive Medicine of Chinese People's Liberation Army. 2016, 34 (4): 464-467.
    Abstract   PDF (3802KB) ( 328 )
Objective To prepare and express the Rta recombinant antigen of Epstein-Barr virus and to establish an ELISA method in order to facilitate the screening of nasopharyngeal carcinoma (NPC).Methods The target gene BRLF1 which coded for Rta was ampliflied by PCR with a specific primer. The recombinant vector pPICZαA-BRLF1 was electronically transformed into pichia pastoris GS115 to express Rta protein so as to screen the His+Mut+ phenotype transformant before proteins were purified and the immunogenicity of the product was proved by Western blot. ELISA was established with the IgG/Rta antibody and was applied to the detection of 300 patients with NPC and 200 healthy people to assess the capability of detection. Results The pPICZαA-BRLF1 recombinant plasmid was successfully constructed and the target protein was secreted and expressed with high-performance (50%). Protein electrophoresis demonstrated a corresponding band in 66 KDa. Western blot showed a good immune-competence with the IgG monoclonal antibody of Rta. The Rta recombinant antigen was used for establishment of the ELISA method. The positive rate of this method was significantly higher than that of IgA/VCA Kit for screening NPC (P<0.01). Conclusion A eukaryotic expression vector of the Rta recombinant antigen of Epstein-Barr virus has been constructed that can ensure the high-performance of foreign proteins with good immunogenicity for the development of IgG/Rta ELISA methods. The detection capability can be improved by combining other antigens and this method is applicable to large-scale screening and diagnosis of NPC.

Journal of Preventive Medicine of Chinese People's Liberation Army. 2016, 34 (4): 468-470.
    Abstract   PDF (3148KB) ( 313 )
Objective To investigate the protective effect of branched-chain amino acid(BCAA)on muscle atrophy in weightlessness-simulated rats. Methods Weightlessness was simulated by a tail-suspended model in rats. SD rats were randomly divided into 3 groups: ground control group (GC), tail-suspended control group (TC) and tail-suspended with BCAA intervention group (TBI).All the rats were successively fed with normal diets. The rats in GC and TC groups received pure water, and those in TBI group were given pure water and intervened with BCAA solution via intragastric administration respectively. The biochemical indices of serum and soleus (SOL) were detected after 4 weeks. Results Compared with GC group, the muscle wet weight(MWW) of and the ratio of MWW to body weight of rats in the TC group were significantly decreased(P<0.01),the level of serum BUN and Ub was significantly increased(P<0.05),the level of serum IGF-1、TESTO、SOD、 and GSH-Px was significantly decreased(P<0.05), and the expression of atrogin-1 protein in SOL decreased by 32%. Compared with TC group, the muscle wet weight(MWW) and the ratio of MWW to body weight were significantly increased (P<0.01) in TBI group,so was the serum level of IGF-1、TESTO、Ub, SOD and GSH-Px,,but the serum BUN and the expression of atrogin-1 protein in SOL were decreased, while the level of SOD and GSH-Px was increased significantly(P<0.05). (P<0.05).Conclusion BCAA may promote the synthesis of SOL protein, but inhibit its breakdown to some extent, alleviating SOL atrophy in tail-suspended rats. The mechanism potentially lies in facilitating the secretion of anabolic hormone and antioxidation ability, and inhibiting the expression of atrogin-1 protein.

    Abstract   PDF (4458KB) ( 343 )
Objective To evaluate the effect of quercetin on axonal growth and functional recovery in mice after spinal cord injury. Methods Twenty-four C57BL/6 female mice were randomly and evenly divided into two groups. Complete compression injury at the spinal cords of T10 was induced in both groups . The mice in therapeutic group were administered with quercetin (50 mg/kg) while control group received the same dose of saline by intraperitoneal injection at 30min before injury, and immediately after injury, and then administered once a day from 1 to 10 days post-injury. The mice in control group received the same dose of saline with an equally long duration. Biotinylated dextran amine (BDA) was injected into the sensorimotor cortex at 14 days post-injury to trace the corticospinal tract (CST). Counterstaining was performed with glial fibrillary acidic protein (GFAP) to label the lesion boundary. The CST retraction and pullulation were observed under a fluorescent microscope. BMS was performed to assess motor functional recovery at 1, 3, 7, 14 and 28 days post-injury.Results Compared with control group, the ratio of total labeled sprouts/axons was significantly increased, the ratio of total labeled retraction bulbs/axons decreased, and the distance of the bundle to lesion border decreased after quercetin treatment at 28 days post-injury. The BMS scores were also significantly improved in therapeutic group (P<0.01).Conclusion Quercetin can promote axonal pullualtion, prevent axonal retraction and improve motor functional recovery after spinal cord injury in mice.

ZHOU Haiyang, WEI Xueming, ZHENG Aimin, DAI Dajiang
Journal of Preventive Medicine of Chinese People's Liberation Army. 2016, 34 (4): 544-546.
    Abstract   PDF (3076KB) ( 319 )
Objective To investigate the association between BRAF mutation and colon cancer with stomach cancer, and between BRAF mutation and gene expression of PRDX6 and PRDX1, and to analyze the role of PRDX1 and PRDX6 gene in colon cancer with other primary cancers. Methods A total of 47 patients with colon cancer complicated by gastric cancer samples were collected. Mutation of BRAF and MLH1 genes were detected by PNAClampTM kit, and chi-square test was used to compare the relationship between mutations and related characteristics. Expressions of PRDX1 and PRDX6 were quantified by qPCR and Western blot in samples with and without BRAF mutation. The plasmid with the BRAF mutation was transferred into human HCT-116 colon cancer cells to investigate the expression levels of PRDX1 and PRDX6.Results BRAF mutation had no correlation with age, cancer location, multiple cancer time intervals and the number of cancer or family history of cancer; while MLH1 mutation had correlation with cancer multiple time intervals and the number of cancer and family cancer history (P<0.05). PRDX1 and PRDX6 expression levels were correlated with the degree of the BRAF mutation and the number of primary cancer in colon cancer (P<0.05). PRDX1 and PRDX6 might be the target of BRAF mutation inhibition. Conclusion BRAF mutation rate in colon cancer complicated by gastric cancer is relatively low in the Chinese population. When using BRAF mutation as a marker for detection, other genes such as MLH1 gene mutation may be as auxiliary. BRAF mutation inhibits PRDX1 and PRDX6, which in turn may affect the number of primary cancer.
