姜黄素对慢性酒精成瘾致肝损伤的干预作用*

Effects of curcumin on liver injury induced by chronic alcohol addiction

摘要: 目的: 探讨姜黄素(Curc)对慢性酒精成瘾性引起的小鼠肝损伤的干预作用。方法: 将30只Balb/c小鼠随机分为正常对照组(Control)、模型组(Model)、低剂量Curc组(5 mg/kg, Curc-L)、中剂量Curc组(10 mg/kg, Curc-M)和高剂量Curc组(15 mg/kg, Curc-H),每组6只。采用20%的白酒制备慢性酒精成瘾性肝损伤模型,Con组每天灌胃2 ml生理盐水;Mod组每天灌胃20%白酒5 ml/kg,Curc治疗组每天用对应浓度的Curc灌胃,每次2 ml,连续35 d。测量小鼠肝脏重量,观察小鼠健康状况。检测小鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)和肝脏中甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、丙二醛(MDA)、总超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮(NO)浓度。观察苏木精和伊红染色的肝组织病理形态学变化。结果: 与对照组相比,模型组小鼠肝脏质量及血清中ALT、AST、ALP、MDA、NO、TC、TG、HDL-C、LDL-C水平显著升高(P<0.05,P<0.01),SOD和GSH-Px活力显著降低(P<0.05,P<0.01),肝细胞有空泡和炎性细胞浸润,肝组织NF-κB和MAPK蛋白表达水平显著上调(P<0.01)。与模型组相比,Curc组的ALT、AST、ALP、MDA、NO、TC、TG、HDL-C、LDL-C的水平显著降低并SOD和GSH-Px活力显著升高(P<0.05,P<0.01)。结论: Curc可能通过调控NF-κB/MAPK信号通路有效地减少肝脏组织损伤。

Abstract: Objective: To investigate the intervention effects of curcumin (Curc) on liver injury induced by chronic alcohol addiction in mice. Methods: Thirty Balb/c mice were randomly divided into normal control group (Control), model group (Model), low-dose Curc group (5 mg/kg, Curc-L), medium dose Curc group (10 mg/kg, Curc-M) and high-dose Curc group (15 mg/kg, Curc-H), with 6 mice in each group. The chronic alcohol addiction liver injury model was prepared with 20% liquor. The mice in control group were given 2 ml of normal saline every day. The mice in model group were given 5 ml/kg of 20% liquor every day, and the mice in Curc treatment group were treated with Curc at the doses of 5, 10, 15 mg/kg in 2 ml saline every day for 35 days. The weight of liver was measured and the health status of mice was observed. Serum ALT, AST, ALP and liver TG, TC, HDL-C, LDL-C, MDA, SOD, GSH-Px and NO were measured. The pathological changes of liver tissues stained with hematoxylin and eosin were observed. Results: Compared with the control group, the liver mass and serum levels of ALT, AST, ALP, MDA, NO, TC, TG, HDL-C, LDL-C in the model group were increased significantly (P<0.05, P<0.01), the activities of SOD and GSH-Px were decreased significantly (P<0.05, P<0.01), the liver cells were vacuolated and infiltrated with inflammatory cells, and the expression levels of NF-κB and MAPK protein in liver tissues were increased significantly (P<0.01). Compared with the model group, the levels of ALT, AST, ALP, MDA, NO, TC, TG, HDL-C, LDL-C in Curc group were decreased significantly nd the activities of SOD and GSH-Px were increased significantly (P<0.05, P<0.01). Conclusion: Curc can effectively reduce liver tissue damage by regulating NF-κB/MAPK signal pathway.
